(P30-019-23) Depletion of Gut Microbiome by Antibiotics Affected the Activity of Dealcoholized Muscadine Wine To Alleviate Dextran Sulfate Sodium (DSS)-Induced Colitis in Mice

Objectives: This study aims to investigate how antibiotics (ABX) affect the activity of dealcoholized muscadine wine (DMW) to alleviate DSS-induced colitis in mice models.

Methods: DMW was prepared by concentrating muscadine wine to 10% of its original volume. Male C57BL/6 mice in the healthy, DSS, and DSS+ABX groups received an AIN-93M diet for 29 days. Mice in DSS+DMW and DSS+ABX+DMW groups received an AIN-93M diet containing 4.19% (w/v) DMW for 29 days. Mice in all four DSS groups received 1.0%-2.5% DSS water on days 19-22 to induce colitis. Mice in two ABX groups received a combined water of ampicillin (1 g/L) and cefazolin (0.47 g/L) on days 1-18 to deplete gut microbiome. Body weight, food intake, stool consistency, rectal bleeding, and disease activity index of mice were recorded every other day. Colon, spleen, and serum were collected after euthanasia for histological and biochemical analysis. Fluorescence of serum was measured 4 hours after oral gavage of fluorescein isothiocyanate (FITC)-dextran to determine intestinal permeability.

Results: A lower concentration of DSS in water induced more severe colitis in ABX-treated mice, suggesting that partial depletion of gut microbiome exacerbated the susceptibility and severity of DSS-induced colitis. DMW reduced weight loss and disease activity index in mice treated with DSS and DSS+ABX (p < 0.05). DMW also reduced the decrease of food intake in DSS+ABX groups (p < 0.05). DMW decreased intestinal permeability by 46.4% and 30.4% in DSS and DSS+ABX mice, respectively (p < 0.05). DMW decreased the spleen weight in DSS+ABX groups by 21.4% but not in DSS groups. DMW decreased the serum secretion of IL-1β by 31.6% in DSS groups but not in DSS+ABX groups. DMW decreased the serum TNF-α level in DSS+ABX mice by 38.7% but not in DSS groups.

Conclusions: Partial depletion of gut microbiome by antibiotics affects the ability of dealcoholized muscadine wine to alleviate DSS-induced colitis in mice.
