(P07-034-23) A Novel Mono- and Diglyceride Emulsion Improves DHA Absorption

Objectives: LLipid absorption is a critical process for healthy growth and development of the human infant. In addition to serving as the primary energy source for the growing infant, lipids are essential for cellular signaling and cell membrane synthesis. Infant pancreatic and hepatobiliary functions continue to mature in the postnatal period, and factors such as preterm birth, GI infection, and nutrition may lead to suboptimal lipid digestion and absorption. Adequate intake of DHA is essential for cognitive and retinal development. This study aimed to assess the capability of a novel mono- and diglyceride and phospholipid emulsion with a hydrolyzed fat blend to improve lipid absorption in a preclinical juvenile rat model.

Methods: Study treatments consisted of a model emulsion containing radiolabeled DHA (³H-DHA). The Control emulsion utilized soy oil, and the experimental emulsion (MDGPL/HF) consisted of a mono- and di-glyceride oil with a hydrolyzed fat blend containing lecithin and cholesterol. Emulsion macronutrients were equalized. Juvenile rats were randomized into two treatment groups, Control (n = 18) and MDGPL/HF (n = 17). Juvenile rats were fasted for 5 hours before receiving 500 µL of either Control or MDGPL/HF emulsion via gavage. Sacrifice and collection of blood and brain tissue were performed at 2-hour intervals following gavage (2, 4, and 6 hr). Lipids were extracted from brain and plasma by the Folch method and measured for radioactivity (dpm).

Results: Rats fed MDGPL/HF emulsion displayed significantly increased plasma ³H-DHA at 6 h (p = 0.028), but not at 2 h (p = 0.20) and 4 h (p = 0.067), compared to rats fed Control emulsion. There was no statistical difference in brain ³H-DHA between the dietary groups, though at 6 h MDGPL/HF displayed a trend (p = 0.112) for increased brain ³H-DHA uptake.

Conclusions: A novel MDGPL/HF emulsion improved acute DHA absorption in a preclinical rat model, and may have application for improving lipid absorption, particularly for pediatric populations. Further study is necessary to assess the effects of both acute and long-term intake MDGPL/HF emulsion intake upon lipid absorption and tissue accumulation.
