(OR10-02-23) Pinto Beans Supplementation Prevents Vascular Dysfunction in an Interleukin-10 Dependent Manner in Female Mice Fed an Atherogenic Diet

Objectives: Risk for cardiovascular disease (CVD) increases in patients with inflammatory bowel conditions, but the underlying mechanism is unclear. Consumption of pinto beans (PB) has been reported to increase ileal expression of the major anti-inflammatory cytokine IL-10. Thus, we investigated the effects of PB supplementation on the inflammatory signature and vascular function of female IL-10 KO mice fed an atherogenic diet.

Methods: Eight-wk-old female B6.129P2-Il10tm1Cgn/J (IL-10KO) and C57BL/6 (WT) were randomized to six groups (n=12/group) in a 2x3 design, with strain (WT or KO) and diets (AIN-93M [CO], atherogenic with 36.5% fat kcal and 1% cholesterol [Ath], or Ath +10% [wt/wt] PB [ PB]). Fecal short-chain fatty acids (SCFAs), ileal T cell populations, estrogen (E2), serum and vascular proinflammatory and proatherogenic markers, as well as markers of vascular endothelial function were assessed after 9 wks. Data were analyzed using 2-way ANOVA. p < 0.05 was considered statistically significant.

Results: PB increased (P < 0.05) fecal acetate and total SCFAs, and prevented the Ath-induced reduction (P < 0.05) in butyrate, but only in WT mice. The expansion of anti-inflammatory T regulatory cells (Tregs) in PB-treated mice was suppressed (~47%) in IL-10 KO mice, while CD4+IL17+ cells only increased (P < 0.0001) in KO mice receiving the CO diet. PB treatment abrogated the increase (P < 0.01) in serum IL-6 and lipopolysaccharide binding protein in both mouse strains receiving Ath. Irrespective of diet, IL-10 KO mice exhibited increased (P < 0.05) serum IL-17A and the proatherogenic marker, trimethylamine-N-oxide (TMAO), but reduced (P < 0.05) cecal β-glucuronidase and serum estradiol. In WT mice, PB rescued the impairment (P < 0.05) in the acetylcholine-induced dilation of the mesentery artery of Ath-fed mice, whereas IL-10 KO impaired this response across all dietary treatments. Increased (P < 0.05) gene expression of macrophage markers, Nos2 and Adgre, in Ath-treated mice was suppressed by PB in WT mice only.

Conclusions: Reduced SCFAs and E2, and an increased TMAO contribute to vascular dysfunction in IL-10 KO mice. PB possesses anti-inflammatory potential and may prevent vascular dysfunction via a mechanism that is IL-10 dependent.
