(P03-032-23) Asperuloside Improves Vascular Function and Fatty Acid Metabolism in Obese Mice

Objectives: Asperuloside (ASP) is shown to be the major active component in Eucommia leaves. This study aims to further explore vascular effects as well as to characterize the molecular mechanism of ASP in obese model.

Methods: After induction of obesity in C57BL/6 male mice fed with high fat diet (60% kcal from fat) for twelve weeks, they were treated daily with ASP (50 mg/kg) via oral gavage for seven weeks. Blood serum was collected for biomarkers detection. Histological examination will be identified by H&E staining. The genes expression levels were detected by real time PCR. Vascular relaxations in aortae were determined by wire myograph. Comparisons among different groups are made using one-way ANOVA followed by Tukey’s test or Two-way ANOVA followed by Tukey’s test for multiple comparison. P values < 0.05 will be considered statistically significant. All results were expressed as mean ± standard error of the mean.

Results: ASP decreased serum cholesterol level (137.9 ± 4.031, n=5, vs 209.1 ± 7.788, n=5, p < 0.0001) and weight of perirenal adipose tissues (2.057 ± 0.1361, n=8, vs 2.569 ± 0.1254, n=8, p=0.03) in obese mice. Meanwhile, ASP ameliorated vascular dysfunction (27.442 ± 4.432, n=6 vs 50.881 ± 3.796, n=6, p< 0.0001) and suppressed elevated ROS level (112 ± 7.475, n=5 vs 210.6 ± 10.19, n=5, p< 0.0001) in obese mice aortae. H&E staining showed ASP inhibited lipid accumulation in obese mice liver. Interestingly, ASP decreased PPARγ (0.8459 ± 0.05, n=5 vs 1.225 ± 0.09, n=5, p=0.01) and CEBPα (0.7361 ± 0.1243, n=5 vs 2.017 ± 0.4587, n=5, p=0.007) in adipose tissues and upregulated SIRT1 (2.306 ± 0.5525, n=7 vs 0.362 ± 0.1716, n=7, p=0.006) and SIRT6 (3.758 ± 0.5678, n=5 vs 1.008 ± 0.4785, n=5, p=0.02) in liver, indicating that ASP may attenuate the early adipogenesis and stimulated fatty acid β-oxidation through activating SIRTs family.

Conclusions: Asperuloside prevented fatty liver and inhibited perirenal adipose tissue expansion, which may through upregulating SIRTs family and attenuating early adipogenesis. Furthermore, we first found that ASP ameliorated obesity associated vascular dysfunction and suppressed vascular oxidative stress.
