(P03-035-23) Effect of Rosmarinic Acid on the Focal Adhesions and Proliferation of Mc3t3-E1 Preosteoblasts on Titanium Surface

Objectives: Focal adhesions (FAs) is the most important process in the first stage of osseointegraction between preosteoblasts and Titanium (Ti). Improving FAs and cell proliferation of pre-osteoblasts leads to successful Ti-based dental implant. Rosmarinic acid (RA) is a natural phenolic substance that have therapeutic potential for diseases.

Methods: MC3T3-E1 preosteoblasts, on Ti discs incubated in serum-free medium for 12 h were incubated with or without 14 µg/ml RA for 10, 30 and 60 min. FAs and cell proliferation were assessed via 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The other experiments were conducted by western blot analysis and immunofluorescence staining.

Results: RA increased FAK phosphorylation, paxillin expression and F-actin formation in MC3T3-E1 cells on Ti surfaces. As a result, RA not only enhances FAs between MC3T3-E1 preosteoblasts and Ti surfaces through the FAK/Paxillin signal pathway, but also increases cell proliferation through the FAK/Grab2/Ras/ERK1/2 signaling pathway.

Conclusions: RA can be applied as an effective functional and therapeutic substance to improve FAs and cell proliferation of MC3T3-E1 preosteoblats on Ti surface, which are essential in the first stage of osseointegration.
