(P03-039-23) A Herbal Blend of Sphaeranthus indicus and Garcinia mangostana Reduces Adiposity in High-fat Diet-induced Obese Mice

Objectives: As the importance of researching a potent but safe anti-obesity treatment is raised, we investigate the effect of Sphaeranthus indicus and Garcinia mangostana, which have been validated for antioxidants and anti-inflammatory properties. This study hypothesized that the herbal blend of these two extracts would suppress fat accumulation in high-fat diet (HFD)-induced obese mice.

Methods: Four-week-old C57BL/6Ntac mice were fed a high-fat diet (HFD, 45% kcal from fat) ad libitum for 20 weeks. Mice were randomly assigned to six groups as follows: 1) normal diet (18% kcal from fat), 2) negative control (45% kcal from fat), 3) low (herbal blend 150 mg/kg bw), 4) medium (herbal blend 300 mg/kg bw), 5) high (herbal blend 450 mg/kg bw), and 6) positive control (metformin 150 mg/kg bw). The herbal blend was supplemented daily by oral gavage. Body weight and feed intake were measured each week.

Results: Body weight was reduced in the high group compared to the negative control (p < 0.05). The herbal blend also significantly decreased body fat mass (p < 0.05). Total feed intake did not show a significant difference among HFD groups. Furthermore, liver weight was reduced by approximately 20% in the medium- and high-dose group (p < 0.05). Similarly to the tendency in body weight changes, the high group significantly reduced serum triglycerides and cholesterol levels (p < 0.001 and p < 0.05, respectively).

Conclusions: The herbal blend of Sphaeranthus indicus and Garcinia mangostana improved body adiposity accumulation. We especially suggest that these herb extracts have a physiological potential for dyslipidemia.
