(P03-082-23) Protective Effect of Mango Consumption Against Metabolic Syndrome in High Sucrose Fed C57BL/6N Mice

Objectives: To evaluate effect of consuming mango products on body weight, blood metabolic parameters and hepatic tissue of high sucrose fed C57BL/6N mice

The greater quantity and variety of bioactive compounds in mango peel will cause a greater protective effect than mango pulp and fiber against metabolic syndrome caused by high sucrose consumption in male mice.

Methods: Male mice of the C57BL/6N strain were fed ad libitum with a standard diet and a 50% sucrose solution for 30 weeks to induce metabolic syndrome, later they were separated into 4 groups (n=6): metabolic syndrome, 10% mango pulp, 10% mango peel and 10% mango peel fiber for 8 weeks, additionally one group fed with standard diet and water was used as a healthy control. Body weight was recorded weekly. Blood and liver tissue were extracted from the euthanized mice, and fast glucose, insulin, HOMA-IR, TG and cholesterol were measured from serum and tissue was fixed in 10% formalin solution, then the tissues were processed in paraffin and stained with hematoxylin and eosin and then assessed according to the published NASH CRN Scoring System.

Results: Mango peel seems to have the major protective effect against metabolic syndrome, since showed minor levels of metabolic indicators HOMA-IR. Besides, mango peel 10% fed mice, presented lower liver tissue of NAS score, mostly in terms of inflammation. This could be related to higher bioactive compounds like mangiferin, quercetin and other polyphenols with proved hepatoprotective activity.

Conclusions: Consumption of mango products caused a positive effect on metabolic syndrome indicators in induced C57BL/6N mice, being the highest effect mango peel consumption followed by mango pulp and peel fiber. Minor body weight and less liver tissue inflammatory infiltrates were the highlight effects in 10% peel fed mice group.
