Objectives: Inflammatory bowel disease (IBD) are complicated multifactorial disease affected by genetic predisposition, dysregulated immune response, and environmental factors. Gut microbiota is recognized as a crucial factor contributing to colitis onset. This study examined the beneficial effects of dietary purple potato on restoring gut microbiota dysbiosis in interleukin (IL)-10-deficient mice, a commonly used IBD mice model.

Methods: IL-10-deficient mice at 7-week-old were assigned to a standard rodent diet (CON) or a control diet supplemented with 10% purple potato (dry feed weight, PP) for 11 weeks, when the fecal samples were collected for Illumina Miseq sequencing and bioinformatic analyses.

Results: Beta diversity of Bray-Curtis distance showed bacterial communities from IL-10-deficient mice with PP supplementation were distinctly separated from CON. At the phylum level, dietary PP intake supplementation decreased Proteobacteria (P < 0.01) and Deferribacteres (P < 0.05), which was closely related to IBD onset. On the other hand, PP increased Firmicutes (P < 0.10), predominant phyla in the microbiota, and was known to be decreased in IBD patients. At the family, PP supplementation bloomed Bifidobacteriaceae (P < 0.01), Porphyromonadaceae (P < 0.01), many species of which are part of intestinal indigenous microbiota, and Lachnospiraceae-Ruminococcaceae family, a major butyrate-producing group. On the contrary, dietary PP supplementation decreased the abundance of Bacteroidaceae, Sutterellaceae, Coriobacteriaceae, Enterobacteriaceae, and Peptostreptococcaceae, all of which were reported to be increased in IBD patients. In addition, dietary PP significantly enhanced Bifidobacterium and butyrate-producing genera while decreasing colitis-associated genera.

Conclusions: Dietary purple potato supplementation ameliorates gut inflammation and colitis likely by promoting beneficial gut microbiota.
