Objectives: Emotional eating that can lead to weight gain is linked to the variations in the genes associated with food craving under stress condition. However, few studies have examined the impact of stress-related genetic polymorphisms on dietary intake and body weight. We investigated the association between the genetic risk level of stress and body weight, and the effect of sleep duration and physical activity on this relationship.

Methods: A total of 100 healthy adults participated in this study. The polygenic risk of stress included DRD2 rs1800497, COMT rs4680, BDNF rs6265, and HRT2C-759C/T genes and was expressed as categorical variables, low (n=29), normal (n=16), alert (n=36), and risk (n=19), or continuous variables of 0-10 scale. Nutrient and food intake was assessed by 3-day dietary record, and physical activity and sleep hour were measured by using Fitbit charge 3. The association between stress-related genetic risk and variables was analyzed by using multivariate linear regression.

Results: The high genetic risk for stress showed a significant association with high BMI (kg/m2) (β=0.195, p=0.028). There was a significant interaction between stress-related genetic risk and sleep hour on BMI (p=0.047). A significant positive association was found between stress-related genetic risk and BMI only in subjects with short sleep duration ( < 6.5 hr/day) when adjusted for sex, age, energy intake, and sleep hour (β=0.726, p=0.038). Furthermore, a significant interaction between stress-related genetic risk and total daily calorie expenditure (kcal/day) on BMI (kg/m2) was observed when adjusted for sex, energy intake, and sleep duration (p=0.025). The stress-related genetic risk was positively associated with the consumption of sugar-sweetened beverages (r=0.355) in subjects with low calorie expenditure ( < 15,000 kcal/wk).

Conclusions: Our findings showed that stress-related genetic risk was associated with BMI, and this association was more pronounced when duration of sleep or physical activity were insufficient. Therefore, individuals with high genetic risk of stress need to be more cautious of emotional overeating and lack of sleep and physical activity.
