Objectives: To investigate Lycium barbarum polysaccharides (LBP) and/or plasmon activated water (PAW) against IFN-γ/TNF-α induced inflammation in Caco-2 cells.

Methods: Caco-2 cells were incubated without treatment (control, C), with IFN-γ/TNF-α induction (NC), with NC pretreated with 100, 250, or 500 μg/mL LBP (L100, L250, or L500), or with the combination of PAW (CP, NCP, L100P, L250P, or L500P). Caco-2 cells (2 x 105) were seeded into wells for 7-10 d, incubated with 10 ng/mL interferon-γ (IFN-γ) for 24 h when the confluency reached >90%, further treated with 100-500 μg/mL LBP for 3 h in serum-free medium prepared by distilled deionized water (DIW) or PAW, and subsequently incubated with 50 ng/mL tumor necrosis factor-α (TNF-α) for 24 h to provoke inflammation. Cells and medium were collected, and levels of interleukin (IL)-6 and IL-8 were assayed. Cell proteins were extracted to determine cycylooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) protein expression using gel electrophoresis and Western blot.

Results: The levels of IL-6 and IL-8 were significantly elevated after the induction of IFN-γ/TNF-α. The levels of IL-6 in the medium were significantly decreased in the L250 group compared to the NC group, and IL-8 levels were tended to be reduced in the L100, L250, or L500 group, but not statistically different compared to the NC group. The combination of LBP and PAW significantly decreased IL-6 or IL-8 compared to the NC or NCP group. The L100P group reduced both IL-6 and IL-8 levels compared to the L100 group. Protein expression of COX-2 and iNOS was significantly increased when the cells were induced with IFN-γ/TNF-α, and the treatment of 100, 250, or 500 μg/mL LBP significantly ameliorated their protein expression. However, no significant differences were found between the groups with or without PAW.

Conclusions: LBP exerted anti-inflammatory potential as it significantly decrease IL-6 levels, ameliorate COX-2 and iNOS protein expression, and combination with PAW shown better effects in against IL-6 and IL-8 secretion, suggesting PAW may contribute synergistic/additive effects with LBP in against inflammatory provoked Caco-2 cells.
