Objectives: Free radicals generated by various endogenous systems in our body cause redox imbalances and can lead to aging and degenerative diseases. Activation of superoxide dismutase (SOD), the first step in the antioxidant system, has been shown to protect the body from oxidative stress by boosting the redox cycle. However, since simple antioxidants research has been the major so far, evaluation of the effectiveness of SOD, an enzymatic antioxidant, is still insufficient. Therefore, the purpose of this study is to evaluate the effects of enzymatic antioxidants through changes in the antioxidant index and circulating microbiome according to long-term intake.

Methods: This study was designed as a randomized, double-blind, placebo-controlled in 80 healthy adults, and subjects consumed SOD (250 IU/capsule) or a placebo daily for eight weeks. Indices of oxidative damage and antioxidant system were evaluated using blood and urine samples collected before and after repeated consumption for eight weeks. Plasma microbiome analysis performed alpha and beta diversity statistics and linear discriminant analysis effect size (LEfSe).

Results: Endogenous antioxidant defense system and oxidative damage indicators were compared by considering the interaction between the group and time. Long-term intake of SOD activated antioxidant enzymes within the test group and remarkably decreased the level of oxidative damage markers, resulting in a difference between groups (P < 0.05). There were no significant differences in microbial alpha and beta diversity between the two groups in Week 8. The linear discriminant analysis effect size (LEfSe) analysis showed that six genera were enriched in the placebo group, while two genera were enriched in the placebo group at Week 8. Then, it was confirmed that there was a correlation between clinical variables and microorganisms at the genus level.

Conclusions: These results suggest that repeated supplementation of SOD can enhance the activity of endogenous antioxidant enzymes and mitigate oxidative damage, potentially related to shifts in microbial composition.
