(P20-049-23) The Necessity To Investigate the Microbiome Member of Pantoea Species in Wheat Grains

Objectives: A new semi-selective medium designated ‘lysine-ornithine-mannitol-arginine-charcoal' (LOMAC) medium to easily isolate Pantoea species(P) in Japan was made with high test efficiency. Pantoea is abundant in plant and animal products, arthropods and other animals, water, soil, dust and air, and occasionally in humans. However, it used hardly characterized the clonality of Pantoea species because of no specific argan medium and no protocol to detect it. Although Pantoea species are known to cause crop diseases etc, they also produce substances effective in the treatment of various cancers in humans and animals, suppress the development　of various plant pathogens via antibiotic production, etc. Therefore, our purpose considers the possibility of Pantoea species by investigating the microbiome member in wheat grains.

Methods: Pantoea species isolated from 5 wheat gains produced by different areas with LOMAC medium. Samples of gains were added to sterile saline and homogenized using a mortar and pestle. After two incubations, bacterial colonies with a yellow color or a orange color were selected. Toneprovided separn stock decided on a nucleic acid sequence using the universal primer of gyrB. We identified a bacterial species using NCBI. 

Results: We gathered 138 strains by LOMAC medium.123 strains of 129 yellow or orange colonies detected Pantea species by PCR. These hosts were corn, onion, fresh rice, buckwheat, tomato, white clover grass, apple, strawberry, and kiwi. The identified species were seven kinds; P.agglomerans etc. While the ratio that a yellow or orange color colonies in wheat grain identified Pantoea species was lower than conventional findings. The identified species were seven kinds in only wheat grain. There were ten kinds of bacteria, including Curtobacterium plantarum except for Pantoea. Clarifying the microbiome in wheat grains suggests improving wheat cultivation methods and nutritional functionality．

Conclusions: Pantoeaspecies is a kind of nitrogen-fixing microbe. O. Maker et al. already published the possibility of including nutrition function by bacterial endophytes in spring wheat grains. We suggest the possibility of alleviating gliadins of the wheat by the cultivating environment with Pantoea species.
