(P22-012-23) Effect of Lifestyle Changes on NLRP3 Inflammasome Expression in Obese Subjects

Objectives: To evaluate the effect of progressive functional exercise and a hypocaloric diet on NLRP3 inflammasome activation  in obese subjects.

Methods: Forty subjects with obesity (BMI >30 kg/m²) with age range of 25-50 years were randomized into two groups: a) hypocaloric diet, or b) hypocaloric diet and progressive functional exercise. The participants were follow up during 3 months. Anthropometric evaluation were performed by bioimpedance (Tanita BC-553), Nutritionist Pro^TM Software was using to evaluated dietary datas. Physical exercise were evaluated by maximum heart rate and Borg scale. Biochemical concentrations were determined by liquid chemistry and insulin levels was performed by ELISA kit. Gene expression of NLRP3 inflammasome complex from peripheral blood (NLRP3, ASC and CASPASA-1 genes) were performed by real-time PCR using TaqMan probe by relative quantification method. Statistical analysis was performed with GraphPad version 9 and SPSS version 25 softwares, 95% confidence interval and p-value < 0.05 were considered as statistically significant.

Results: Both group significant decrease anthropometric variables (BMI, weight, fat mass, body water, muscle mass, and waist circumference). Beside, might be a significant decrease on NLRP3 inflammasome complex in the intervention group compared to the control group.

Conclusions: Lifestyle changes such as hypocaloric diet and progressive functional physical exercise regulate the expression of the NLRP3 inflammasome, which is a complex associated with the development of chronic diseases related to obesity, that are the main causes of death in worldwide.
