Objectives: Lycopene has strong antioxidant capacity and has been discussed as a potential effector in the prevention and therapy of cardiac disorders. Phthalates as a large group of environmental pollutants have been strongly correlated to the onset of cardiac disorders. The purpose of the present research was to determine the mechanism of lycopene preventing persistent pollutant induced cardiac disorders in mice.

Methods: ICR mice were treated by gavage with water, corn oil, 500mg/kg DEHP B.W. and/or 5 mg/kg B.W. LYC for 28 days. Thenceforth, additional potential mechanisms of lycopene preventing persistent pollutant induced cardiac disorders was investigated.

Results: Our research showed that LYC improved DEHP-induced histopathological changes involving cardiomyocyte lysis and rupture, and ultrastructural damage such as dissolution and loss of mitochondrial cristae. Furthermore, lycopene alleviated DEHP-induced oxidative stress and significant decline in the antioxidant function, which inhibited the activation of nuclear factor E2-related factor 2 (Nrf2)/ heme oxygen-1 (HO-1) signaling pathway. Interestingly, lycopene reversed DEHP resulted lipid peroxidation and increased ferrous ion content, suggesting that ferroptosis occurred in mouse cardiac.

Conclusions: Overall, our findings demonstrated that lycopene could relieve DEHP-induced cardiac disorders via downregulation of HO-1. The present study provides novel evidence of HO-1 as a target for lycopene preventing persistent pollutant induced cardiac disorders.
