(P23-004-23) Effects of a Combination of Dietary Resistant Starch and Low Dose Captopril on Metabolic Syndrome Biomarkers in a Type 2 Diabetes Rodent Model

Objectives: To evaluate a potential combinatorial approach using dietary resistant starch (RS) and a renin-angiotensin system inhibitor (captopril), on key metabolic syndrome biomarkers in a type 2 Diabetes rodent model.

Methods: Male Zucker diabetic fatty rats (ZDF, 6 wk) and lean Zucker rats (n=8/group), were fed diets containing either 0%, 5%, or 10% RS, and injected daily with a low captopril dose (25mg/kg, body weight) or vehicle (saline) injections; Animals were put in metabolic cages 12 h. before being anesthetized by intraperitoneal injection with a cocktail of ketamine: xylazine (90:10 mg/kg body) after which whole blood was collected by cardiac puncture and analyzed for fasting blood glucose (FBG). Serum insulin and triglycerides (TG’s) were analyzed in serum using commercial kits. Data were analyzed using Mann-Whitney tests.

Results: Groups taking the captopril treatment, regardless of the RS content assigned, displayed attenuated FBG values around 61-69% compared to the ZDF 0%RS+placebo group, tentatively indicating a regulating effect of captopril on this biomarker. Data showed that groups receiving captopril injections showed average TG’s values (mg/dl) of 211.2±50.4, 236.9±71.2, 282± 79.6 for 0%, 5%, 10% RS respectively, which were 60-70% lower than those of their counterparts that took placebo injections. Groups fed RS diets didn’t exhibit any apparent improvements in the insulin resistance indicators HOMA IR and HOMA β regardless of treatment with captopril.

Conclusions: Captopril attenuated FBG levels in all groups, and reduced serum TG concentrations compared to the vehicle-treated (placebo) groups. Overall insulin resistance indicators were not conclusive on whether RS or captopril modulated insulin concentrations.
