(P23-018-23) Dealcoholized Muscadine Wine Improved Skin Health, Reduced Oxidative Stress and Inflammation in Women in a Randomized Controlled Trial

Objectives: Skin aging is caused by inflammation, oxidative stress, and UV radiation. This research aimed to determine the effect of dealcoholized muscadine wine (DMW) on skin aging and UVB-induced erythema in healthy females aged 40-67 using a randomized, single-blinded, placebo-controlled, cross-over design.

Methods: The research was approved by IRB and registered on Clinicialtrials.gov (NCT04614753). Seventeen healthy women aged 40-67 with Fitzpatrick skin types 2 and 3 completed this study. Participants were advised to avoid Vaccinium products, botanical dietary supplements, and probiotic supplements during the study. After a 7-day run-in period, participants consumed 300 mL of DMW or a placebo for six weeks. Following a 21-day wash-out period, participants switch to the opposite beverage for additional 6 weeks. Skin conditions were measured at baseline and after six weeks, including transepidermal water loss, elasticity, UVB-induced erythema, and others. Inflammatory and oxidative stress biomarkers in the blood were assessed using ELISA and colorimetric methods. A repeated-measures mixed model was used to determine statistical significance of the measurements.

Results: DMW intake significantly decreased transepidermal water loss on the face from 15.0 to 11.2 g/h/m² (p≤0.05) compared to baseline. Skin pH was significantly decreased while net elasticity was significantly increased by DMW compared to baseline (p≤0.05). Gross elasticity significantly increased after DMW consumption compared to the placebo (p≤0.05). On the forearm, DMW consumption increased gross elasticity and net elasticity, while decreasing viscoelasticity compared to baseline and placebo (p≤0.05). Smoothness was increased and roughness was decreased compared to baseline (p≤0.05). In the blood, glutathione peroxidase activity increased, while the levels of advanced glycation end products and matrix metalloprotease-9 were decreased by DMW consumption (p≤0.05) compared to baseline.

Conclusions: This randomized controlled trial demonstrated that consumption of polyphenol-rich DMW improved skin conditions associated with aging. This is likely due to decreases in inflammation and oxidative stress.
