Objectives: Use the targeted metabolomic analyses of plasma to explore the effect of 8-week different dose of Bifidobacterium supplementation combined with training on the metabolism of cross-country skiers.

Methods: Sixteen male cross-country skiers who volunteered to participate in the experiment were recruited and randomly divided into low dose group (L, count 1*108CFU) and high dose group (H, count <1*109CFU). Supplementary scheme is 4 times/day (with the three meals and at 21:00 in the evening), 6 days/week, for eight weeks. The experiment was conducted in a double-blind mode. The dietary and energy consumption of the subjects were recorded every week. Blood was collected before and after the experiment. Body composition, maximal oxygen consumption (VO2 max), blood biochemical test and blood targeted metabolomics were measured.

Results: (1) After 8 weeks, there are no significant difference of the dietary intake, energy consumption, body composition and VO2 max both in L and H group. (2)Triglyceride and LDL-C content of both L (0.6±0.1 vs 0.3±0.0 mmol/L, P＜0.05) and H (0.7±0.1 vs 0.4±0.0 mmol/L, P＜0.05) group were significantly decreased. (3) Both in L and H group, the enrichment results of differential metabolite pathway include the biosynthesis of polyunsaturated fatty acids. (4) Metabolites of fatty acids in blood including acetoacetic acid and four kinds unsaturated fatty acids (5-Dodecenoic acid, 10Z-Heptadecenoic acid, Adrenic acid, 8-11-14-Eicosatrienoic acid) were significantly increased more than 1.5 times, while the blood content of Glycocholic acid related to fat digestion and absorption was significantly decreased 3 times. (5) Correlation analysis find valeric acid (mainly produced by the gut microbiota) is negatively correlated with TG and HDL-C in blood. Unfortunately, there was no significant difference in all indicators between L and H Group.

Conclusions: 8-week Bifidobacterium supplementation with training may help cross-country skiers improve fat metabolism by increasing fat catabolism and reducing fat absorption.
