Objectives: Our group previously reported the anti-inflammatory effects of a phenolic-enriched maple syrup extract (MSX) in murine macrophages by reducing the levels of nitric oxide and prostaglandin-E2 and inhibiting the transcriptional activation of nuclear factor-κB. In addition, our preliminary in vivo experiments showed that MSX can reduce the levels of pro-inflammatory cytokines such as Interleukin 1 (IL-1) in a diet-induced mouse diabetes model. However, the effect of MSX on the activation of inflammasome, which is a critical molecular target regulating the production of pro-inflammatory cytokines including IL-1, is unknown. Herein, we evaluated MSX’s inhibitory effect on inflammasome activation in phorbol 12-myristate 13-acetate differentiated human monocytes THP-1 cells.

Methods: A cellular inflammasome model was established by stimulating THP-1 cells using lipopolysaccharide (LPS; 100 ng/mL) and nigericin (10 μM).

Results: The stimulation with LPS and nigericin elevated the production of inflammatory cytokines including IL-1β and IL-6 to 530.6 and 975.2 pg/mL, respectively, as compared to the control group. Treatment with MSX at concentrations of 1, 10, and 100 μg/mL showed anti-inflammatory effects by reducing the level of IL-6 induced by the stimulation of LPS+nigericin to 971.6, 829.2, and 653.8 pg/mL, respectively. In addition, treatment with MSX at concentrations of 1, 10, and 100 μg/mL did not significantly alter the production of IL-1β induced by LPS+nigericin, as evidenced by the levels of IL-1β at 700.3, 761.9, and 715.2 pg/mL, respectively, in the MSX-treated cells. Furthermore, we evaluated the modulatory effects of MSX on inflammasome-related targets (e.g. caspase-1) and pathways (e.g. pyroptosis) in THP-1 cells, which elucidated the underlying mechanism of MSX’s anti-inflammasome effects.

Conclusions: Findings from the current study provide additional insights into the anti-inflammatory effects of MSX in the context of IL-6 production, which support the potential nutraceutical applications of MSX.
